Subject: PulseNet Quick Tip: PFGE Equipment Maintenance Series, Replacing Electrodes

1.       Disconnect the power supply and tubing from the electrophoresis chamber.  Turn over the electrophoresis chamber, unscrew the feet, and remove the screws holding the base plate with a Philips screwdriver.  Remove the base plate.

2.       Remove the hexagonal nut on the wire, the wire, second nut, and washer.  Multiple electrodes can be changed simultaneously.  It is important to keep track of the wire for each associated electrode if multiple electrodes will be replaced at one time.            

3.       Push down firmly on the post of the broken electrode to remove it.

4.       Insert new electrode from front side of chamber so that post of electrode appears through opening.  Carefully fill the opening around the post with self-leveling silicone sealant and allow the sealant to cure for at least 24 hours.

5.       Replace the washer, hexagonal nut, wire, and second nut. Replace the base plate/feet and reconnect the tubing/power supply.
